ÉTUDE DES INTERACTIONS DES MÉLANGES.(2017-12-11) MERINE HANANE; Encadreur: MERINE HAOUARIA; Co-Encadreur: ELBAHRI ZinebRésumé (Français et/ou Anglais) : Résumé L’optimisation de l’incorporation et de la libération des principes actifs dans les produits formulés constitue un des enjeux majeurs des industries pharmaceutiques et cosmétiques. L'objectif principal de ce travail est l’étudede la libération des principes actifs suivants: l’Antipyrine, laKetoprofen et la Benzocaïne, à partir de différentes formes à base depolymères biodégradableset biocompatibles(PCL et PLA) ainsi que leursinteractions. Deux types de systèmes matriciels ont été développés:disques à libération prolongée réalisés par simple dispersion du principe actif dans la matrice polymères et microsphères obtenues par la méthode de microencapsulation par émulsion-évaporation de solvant. Le suivi in vitro de la libération des principes actifs, réalisé dans des milieux physiologiques reconstitués de pH gastro-intestinaux, nous a permis d’étudier d’une part l’effet de la nature du polymère ou du copolymère matrice et de sa masse moléculaire et d’autre part l’effet du procédé de formulation sur le profil et la vitesse de libération.Enfin, des modèles théoriques et empiriques ont été testés pour élucider le mécanisme et prédire le profil de libération des principes actifs choisis. Abstract The optimization of the drug incorporation and the drug release constitutes one of the major stakes in the pharmaceutical and cosmetic industries. The main objective of this work is to study the release of the following active ingredients: Antipyrine, Ketoprofen and Procaine from different dosage forms based on biodegradable and biocompatible polymers (PCL and PLA) and theirs interactions. Two types of matrix systems were developed: disks for sustained release obtained by dispersion of the active ingredient in the matrix and microspheres elaborated by the microencapsulation process based on the emulsion-solvent evaporation. The in vitro drug release established in gastro-intestinal fluids pH permitted to study,on the one hand the effect of the nature of the polymer or the copolymer matrix and its molecular weight and on the other hand the effect the process formulation both on the release profile and the release rate. Finally, theoretical and empirical models were tested in order to elucidate the release mechanism and to predict the release profile of the selected actives ingredients.
