    Contribution à l’étude phytochimique et valorisation de l’espèce Agave americana L. de l’Ouest algérien

    Résumé (Français et/ou Anglais) : L’objectif de notre recherche est d’évaluer les activités biologiques des extraits de l'Agave americana L en provenance de la région de Kristel et Tessala. Le screening phytochimique effectué sur les différents extraits de l'Agaveamericanarévèle la présence de : phénols, flavonoïdes, tanins, terpènes ainsi que les saponines et l'absence des alcaloïdes. L'étude phytochimique a révélé que l'extrait de décoction des feuilles de la région de Kristel renferme les teneurs les plus élevées en phénols totaux (62,44±0.48 mg AGE/g MS), de flavonoïdes (8.385± 0.01mg EC/g MS) et de tanins condensés (3.63±0.27 mg EC/g MS). Tandis que, les extraits hydro-alcooliques enregistrent les valeurs les plus notables en composés phénoliques pour les racines. La fraction de chloroforme des deux régions enregistre la quantité la plus importante en phénols totaux avec une teneur de (162.82±14.30 mg AGE/100g MS – 141±8.19 mg AGE/100g MS). Tandis que, les valeurs les plus élevées en flavonoïdes ont été obtenu pour l'extrait de chloroforme et acétate d'éthyle de Kristel (34,32 ± 3,31 mg EC / 100g Ms; 31,17 ± 3,05 mg EC/ 100g Ms). L'évaluation de l'activité antioxydant par le test DPPH, montre que tous les extraits possèdent une activité anti-radicalaire importante. La fraction d'acétate d'éthyle et la décoction de la région de Kristel renferme le pouvoir antioxydant le plus remarquable avec respectivement une concentration d'inhibition IC50 de 1.30 mg/ml et 2.23 mg/ml. L'étude in vivo de l'induction de l'œdème par la carragénine, chez les rats Wistar des extraits hydro-éthanolique (EEAA) et de décoction (EDAA), en comparaison avec l'indométacine et le piroxicam montre que les deux extraits possèdent un effet anti-inflammatoire très intéressant et ils agissent dès la première heure. L’EEAA renferme un effet anti-inflammatoire plus puissant que l'EDAA, donc l'effet anti-inflammatoire dépend de la nature chimique des molécules bioactives présentent dans les extraits. La caractérisation du profil chimique des différentes fractions des deux régions, par la technique de chromatographie liquide à haute performance a identifié cinq composés flavonoïdiques, dont cinq pour la région de Kristel (Naringine, Taxifoline, Catéchine, Apéginine, Fiestine), et une dans la fraction d'acétate d'éthyle (Naringine) pour la région de Tessala. The objective of our research is to evaluate the biological activities of the extracts of Agave americana L., from the region of Kristel and Tessala. The phytochemical screening carried out on the various extracts of Agave americana reveals the presence of: phenols, flavonoids, tannins, terpenes as well as saponins and the absence of alkaloids. The phytochemical study revealed that the decoction extract from the Kristel region contained the highest total phenols (62.44 ± 0.48 mg GAE / g MS), flavonoids (8.385 ± 0.01 mg CE / g MS) and condensed tannins (3.63 ± 0.27).While, hydro-alcoholic extracts record the most notable values of phenolic compounds for roots. The chloroform fraction of the two regions recorded the most remarkable amount of total phenols with a content of (162.82 ± 14.30 mg GAE / 100 g MS - 141 ± 8.19 mg GAE / 100 g MS). Whereas, the highest flavonoid values were obtained for Kristel's chloroform and ethyl acetate extract (34.32 ± 3.31 mg CE / 100 g Ms, 31.17 ± 3.05 mg CE / 100g Ms). Evaluation of the antioxidant activity by the DPPH test shows that all the extracts possess a remarkable anti-radical activity. The ethyl acetate fraction and decoction of the Kristel region contained the most remarkable antioxidant power with an IC50 inhibition concentration of 1.30 mg / ml and 2.23 mg / ml, respectively. The in vivo study of carrageenan edema in Wistar rats of the hydroethanolic extract and the decoction extracts in comparison with indomethacin and piroxicam shows that both extracts have a very remarkable anti-inflammatory effect and they act from the first hour. EEAA has a stronger anti-inflammatory effect than EDAA, so the anti-inflammatory effect depends on the chemical nature of the bioactive molecules present in the extracts. The characterization of the chemical profile of the different fractions of the two regions by the high-performance liquid chromatography technique identified five flavonoid compounds, including five for the Kristel region (Naringine, Taxifoline, Catechin, apeginine, Fiestine) and one in the Fraction of ethyl acetate (Naringine) for the Tessala region.
