- ItemLIBERATION CONTROLEE DE L’ACIDE SALICYLIQUE ET L‘ESOMEPRAZOLE A PARTIR DE DIFFERENTES FORMULATIONS(2021-07-18) MEHIDA KHEIRA; Encadreur: MERINE HAOUARIAالملخص المواد المساعدة مثل البوليميرات تستعمل لإنتاج الأدویة المتمیزة بالتحریر الممددلعنصرها الفعال . يستعمل مبدأ التحرير الممدد للتقليل من مقدار الأخذة اليومية للدواء، و كذا تنظيم و إبقاء نسبة من العنصر الفعال تسير في الدم على مدى كل اليوم و بالتالي التموين بفعالية أفضل الهدف من هذه الدراسة هو إنتاج صيغ متميزة بالتحرير الممدد لعنصرها الفعال و المتعلقة ب ايزوميبرازول و حمض ساليسيليك ابتداءا من أشكال مختلفة على قاعدة بوليميرية حيوية قابلة للتحلل و كذلك دراسة تفاعلاتهم ثم تطوير نوعين من نظام المصفوفات : الأقراص ذات التحرير المستمر المحضرة عن طريق وضع المكون النشط داخل مصفوفة البوليمير و الميكروسفار المتحصلعليهما عن طريق تبخر المذيبات المراقبة المخبرية لتحرير المكونات النشطة حضرت في وسط فيزيولوجي للسوائل المعوية و التي تمكننا بدراسة من ناحية تأثير طبيعة مصفوفة البوليمير أو الكوبوليمير و كتلته المولية , و من ناحية أخرى تأثير نظام التغليف على سرعة التحرير أخيرا تم اختبار نماذج نظرية و تجريبية من اجل توضيح آلية التحرير و التنبؤ بخصائص تحرير المكونات النشطة المختارة :الكلمات المفتاحية أقراص , ميكروسفير , البوليميرات الحيوية القابلة للتحلل , التحرير المستمر , نموذج الانتشار , تأثير التأخير Résumé: Des excipients comme les polymères sont utilisées pour formuler des spécialités pharmaceutiques présentant des cinétiques de libération prolongée du principe actif. Ce principe de libération est utilisé pour diminuer la fréquence des prises quotidiennes, réguler et maintenir un taux circulant dans le sang de principe actif tout au long de la journée et donc de fournir une meilleure efficacité. Le but du présent travail a été de concevoir des formulations à libération prolongée de l’acide salicylique et de l’ésomeprazole et étudier leurs libérations à partir de différentes formes à base de polymèresbiodégradableset biocompatibles (PCL, EC et ECg) ainsi que leurs interactions. Deux types de systèmes matriciels ont été développés : disques à libération prolongées réalisées par simple dispersion du principe actif dans la matrice polymère et microsphère obtenues par la méthode de micro encapsulation par émulsion –évaporation de solvant. Le suivi in vitro de la libération des principes actifs, réalisé dans des milieux physiologiques reconstitués dePH gastro-intestinaux nous a permis d’étudier d’une part l’effet de la nature du polymère ou du copolymère matrice et de sa masse moléculaire et d’autre part l’effet du procédé de formulation sur le profil et la vitesse de libération. Enfin des modèles théoriques et empiriques ont été testés pour étudier le mécanisme et prédire le profil de libération des principes actifs choisis. Abstract: Abstract: Excipients like polymers are used to formulate medical products presenting the kinetics of release prolonged of the active ingredient. This principle of release is used to decrease the frequency of the daily catches, to control and maintain a rate circulating in the blood of active ingredient throughout the day and thus to provide a better effectiveness. The goal of this work was to conceive formulations with release prolonged of acid salicylique and ésomeprazole from different dosage forms based on biodegradable and biocompatible polymers (PCL, EC et ECg) and theirs interactions. Two types of matrix systems were developed: disks for sustained release obtained by dispersion of the active ingredient in the matrix and microspheres elaborated by the microencapsulation process based on the emulsion-solvent evaporation. The in vitro drug release established in gastro-intestinal fluids PH permitted to study. On the one hand, the effect of the nature of the polymer or the copolymer matrix and its molecular weight and the other hand the effect the process formulation both on the release profile and to predict the release profile of the selected actives ingredients. L'objectif principal de ce travail est l’étudede la libération des principes actifs suivants: l’Antipyrine, laKetoprofen et la Benzocaïne, à partir de différentes formes à base depolymères biodégradableset biocompatibles(PCL et PLA) ainsi que leursinteractions. Deux types de systèmes matriciels ont été développés:disques à libération prolongée réalisés par simple dispersion du principe actif dans la matrice polymères et microsphères obtenues par la méthode de microencapsulation par émulsion-évaporation de solvant. Abstract The optimization of the drug incorporation and the drug release constitutes one of the major stakes in the pharmaceutical and cosmetic industries. The main objective of this work is to study the release of the following active ingredients: Antipyrine, Ketoprofen and Procaine from different dosage forms based on biodegradable and biocompatible polymers (PCL and PLA) and theirs interactions. Two types of matrix systems were developed: disks for sustained release obtained by dispersion of the active ingredient in the matrix and microspheres elaborated by the microencapsulation process based on the emulsion-solvent evaporation. The in vitro drug release established in gastro-intestinal fluids pH permitted to study,on the one hand the effect of the nature of the polymer or the copolymer matrix and its molecular weight and on the other hand the effect the process formulation both on the release profile and the release rate. Finally, theoretical and empirical models were tested in order to elucidate the release mechanism and to predict the release profile of the selected actives ingredients.
